Figure 7. Xnf7 is required for the spindle assembly checkpoint in egg extracts. CSF extracts were preincubated with preimmune IgG or anti-Xnf7 antibodies for 15 min at 23°C. Demembranated sperm chromatin (9,000/μl) and nocodazole (10 μg/ml) were added for 30 min, followed by the addition of CaCl2. Aliquots removed at the indicated times after CaCl2 addition were analyzed by SDS-PAGE and immunoblotting with anti–cyclin B2 antibodies. White line indicates samples run on the same gel were rearranged. Aliquots were also removed and visualized by Hoechst 33258 staining. Photographs of nuclear morphology at 60 min are shown.
