FIGURE 2 Injections of TALEN or CRISPR/Cas9 targeting tumor suppressor genes lead to tumorigenesis in Xenopus tropicalis, whilst
targeted injections culminate in increased tissue restrictiveness of tumor development. (a) Depending on which stage and in which
blastomere(s) the X. tropicalis embryo is injected with CRISPR/Cas9 targeting rb1 and rbl1, it will exhibit variable levels of genome editing
mosaicity. As such, tadpoles develop retinoblastoma either unilaterally or bilaterally. With targeted injection, it is possible to get tissuerestricted
CRISPR/Cas9 targeting, which reduces the likelihood of embryo-lethality associated with knock-outs (Clarke et al., 1992; Moser
et al., 1995). (b) Targeting apc in the 8-cell dorsal-vegetal blastomere leads to development of exclusively desmoid tumors (red arrows). (c)

et al., 2016). (d) Development of small cell lung cancer in a rb1 and rbl1 CRISPR/Cas9 targeted embryo Image published in: Naert T et al. (2017) Copyright © 2017. Image reproduced with permission of the Publisher, John Wiley & Sons. Larger Image Printer Friendly View |
