Constructs encoding EGFP alone or wild type XDppa2/4 were transfected as controls. (B) Only XDppa2/4 proteins with an intact SAP domain reveal chromatin association as shown by co-staining with histone H2B-mRuby (red) and DAPI (blue). (C) Results of subcellular localization experiments in comparison with phenotypes obtained after overexpression of the respective deletion constructs. A diffuse localization within the nucleus was interpreted as indication that the respective mutant is not associated with chromatin.
