Fig. 3. In situ hybridization analysis of fullback RNA expression at gastrula and neurula stages. In situ hybridization was performed on intact (A,C,E,H) or bisected (B,D,F,G) albino Xenopus embryos. Dashed lines in panel (E) indicates the plane in which the embryo in (F) was cut in half. (A) Vegetal view of an early gastrula (stage 10). (B) Bisected embryo at early gastrula stage 10. (C) Posterior view of an embryo at mid-gastrula stage 11.5. (D) Embryo bisected sagittally at mid-gastrula stage 11.5. Note staining in involuted mesoderm. (E) Dorsal view of an embryo at early neurula stage 13. Arrows indicates stripes of staining on the dorsal side and dashed line indicates plane of section of embryo in (F). (F) Bisected embryo at early neurula stage 13. (G) Embryos bisected sagittally at early neurula stage 13. Note posterior staining. (H) Dorsal view of an embryo at late neurula stage 18. For all images, Vg, vegetal; D, dorsal; V, ventral; A, anterior; P, posterior.

Fig. 4. In situ hybridization analysis of fullback and muscle-specific actin at neurula and tailbud stages. (A,B) Embryos at late neurula stage 18 were bisected transversely and the resulting halves were probed with a fullback (A) or muscle-specific actin (B) probe. Note that the expression domains of fullback and muscle-specific actin overlap to a very limited extent in the dorsal region of the embryo. (C). Intact embryo at stage 24 showing expression of fullback in somites and in a stream of cells which derive from the hindbrain (arrowhead). (D,E) Embryos at stage 25 were bisected transversely and the resulting halves were probed with a fullback (D) or muscle-specific actin (E) probe. Note that the expression domains of fullback and muscle-specific actin overlap in the dorsal regions of the somites. (F) Embryo at stage 35. Note the expression of fullback in the cranial nerves (white arrows), branchial arches (black arrows), diencephalon (d) and midbrain (m). Similar expression patterns to those observed in Fig. 3 and Fig. 4 were obtained with fullback-like antisense probe and with probes derived from the 3′ untranslated regions of fullback and fullback-like.
