The cdk8 protein is a cyclin-dependent kinase and a component of the Mediator complex. It localizes to the nucleus where it phosphorylates the carboxy-terminal domain (CTD) of RNA polymerase II. This activity represses transcription initiation by inhibiting the formation of the pre-initiation complex. Its expression is detected in oocytes and throughout early embryonic development, including stages of gastrulation and neurulation. The protein interacts with cyclin C to form a kinase module that can reversibly associate with the core Mediator complex to regulate gene expression. [-]
