Figure 6 XenMARK annotation of in situ hybridization patterns from both whole tadpoles and retinal sections. Illustration of the annotation process of whole mount (A,B) or retinal (D,E) expression patterns using the XenMARK bioinformatic tool. The procedure consists in manually transferring the expression profiles observed on original images to schematic diagrams, by direct mark-up of selected zones. Intensity of the staining can be distinguished. In (B) for instance, strong expression in the brain and eye is drawn in green, while faint expression in the spinal cord is in red. D,E: Show an example of a complex retinal expression pattern (D), where three sites of expression (arrows) have been drawn to the schematic (green in E). C,F: The full set of annotations is superimposed on heatmap diagrams, revealing the lowest to densest caches of data. Heatmap color scales reflect numbers of annotated images at each pixel. Of note, pixels outside the retina schematic correspond to the corneal epithelium on one hand, and the periocular mesenchym on the other hand, where a couple of clones were expressed. Scale bar = 500 um (A) or 50 um (D). Color figure can be viewed in the online issue, which is available at wileyonlinelibrary.com.
