Figure 3. ZC4H2 inhibits myogenic differentiation of C2C12 cells. (a–c) mRNA levels of ZC4H2, myogenic markers MyHC and Myf5 during myogenic induction in C2C12 cells with ZC4H2 over-expressed. C2C12 cells were firstly transfected with the indicated plasmids 8 h before myogenic induction in DMEM with the addition of 2% horse serum for 2 days. After induction, total RNAs were isolated and analysed by real-time PCR. (d–f) mRNA levels of ZC4H2, myogenic markers MyHC and Myf5 during myogenic induction when ZC4H2 was knocked-down. C2C12 cells were transfected with the indicated siRNAs and then treated with the induction medium for 2 days.
