HIPK2 inhibits axis duplication induced by the injection of β-catenin mRNA into Xenopus embryos. (A) In vitro transcribed β-catenin mRNAs were injected into Xenopus embryos together with either HIPK2 or HIPK2(K221R) mRNA. Note that co-injection of HIPK2 mRNA (500 pg) with β-catenin mRNA (250 pg) resulted in complete or partial inhibition of axis duplication. Typical images of dual axis and partial dual axis embryo are shown. Arrows indicate duplicated axis. (B) Inhibition of axis duplication is quantitatively presented by percentage of embryo. (C) RNAs were isolated from the Xenopus embryos shown in (A), and expressions of Wnt/β-catenin target genes such as Siamois, Xnr3, xCyclin D and Goosecoid (Gsc) were determined by RT-PCR.
