All constructs displayed hallmark characteristics of WT CFTR including requirement for phosphorylation by PKA (here 300 nM) and exposure to MgATP (here 1 mM for WT, 2 mM for others) for channel activity, slow burst kinetics, and locking in the open state by a mixture of ATP (0.1 mM) and PPi (2 mM).
